Abstract

õ¿¬ÀÇ ´ç¾ËÄÝ°è °¨¹Ì·áÀÎ ÀÚÀϸ®ÅçÀ» »ç¿ëÇÏ¿© ÀÚ´çÀ¸·ÎºÎÅÍ ±Û·çĵÀ» ÇÕ¼ºÇÏ´Â È¿¼ÒÀÎ glucosyltransferase(GTF)ÀÇ mRNA ¹ßÇö¿¡ ¹ÌÄ¡´Â ¿µÇâÀ» fluorescent in situ hybridization(FISH)°ú À¯½Ä¼¼Æ÷ÃøÁ¤±â¸¦ ÀÌ¿ëÇÏ¿© ºÐ¼®ÇÏ¿© ´ÙÀ½ÀÇ °á°ú¸¦ ¾ò¾ú´Ù.
1. FISH ºÐ¼®°á°ú 1% ÀÚ´çÀÌ ÇÔÀ¯µÈ BHI ¹èÁö¿¡ 1%, 5%, 10% ÀÚÀϸ®ÅçÀ» ÷°¡ÇÏ¿´À» ¶§ gtfB, gtfC ¹× gtfD mRNA, ¹ßÇöÀÌ ³óµµ ÀÇÁ¸ÀûÀ¸·Î ¾ïÁ¦µÇ¾ú°í, gtfB º¸´Ù´Â gtfC ¹× gtfDÀÇ ¹ßÇöÀÌ ´õ Å©°Ô ¾ïÁ¦µÇ¾ú´Ù.
2. À¯½Ä¼¼Æ÷ÃøÁ¤±â·Î ºÐÇÑ °á°ú ÀÚ´çÀÇ ³óµµ°¡ 0.1%, 0.5%, 10%·Î Áõ°¡ÇÔ¿¡ µû¶ó gtfB, gtfC ¹× gtfD mRNA ¹ßÇöÀÌ Áõ°¡ÇÏ¿´´Ù.
3. À¯½Ä¼¼Æ÷ÃøÁ¤±â·Î ºÐ¼®ÇÑ °á°ú ÀÚ´çÀÌ 1% ÇÔÀ¯µÇ¾úÀ» ¶§ 1%, 5%, 10% ÀÚÀϸ®ÅçÀ» ÷°¡ÇÑ °æ¿ì gtfB, gtfC ¹× gtfD mRNA ¹ßÇöÀÌ À¯»çÇÏ°Ô ¾ïÁ¦µÇ¾úÀ¸¸ç, 10%ÀÚÀϸ®ÅçÀ» ÷°¡ÇÑ °æ¿ì gtfB, gtfC ¹× gtfD mRNA ¹ßÇöÀÌ °¡Àå Å©°Ô ¾ïÁ¦µÇ¾ú´Ù.
ÀÌ»óÀÇ °á°ú¸¦ Á¾ÇÕÇϸé ÀÚÀϸ®ÅçÀº ÀÚ´ç÷°¡¿¡ ÀÇÇÑ Streptococcus mutansÀÇ gtf mRNA ¹ßÇöÀ» ¾ïÁ¦ÇÏ¿© Ç׿ì½ÄÈ¿°ú¿¡ ±â¿©ÇÏ´Â °ÍÀ¸·Î »ý°¢µÈ´Ù.
Xylitol, a five-carbon natural sugar alcohol, is widely used non-cariogenic sugar substitute. In present study, the effects of xylitol on the expression of mRNA for glucosyltransferase which synthesizes glucan from sucrose were detected by Fluorescent in situ hybridization (FISH) and flow cytometry. FITC fluorescences for mRNA of gtfB, gtfC and gtfD were decreased further with increasing concentration of xylitol from 1% to 10% when detect¡þed by FISH. Flow cytometric analysis also showed that the expression of gtfB, gtlC and gtfD was increased by the addition of sucrose and decreased by the addition of xylitol to BHI broth containing 1% sucrose. In conclu¡þsion, the expression of gtfB, gtIC and gtfD mRNA was decreased by the addition of xylitol.
